Patch-Clamp Applications and Protocols By Richard A. Levis, James L. Rae (auth.), Alan A. Boulton, Glen B. Baker, Wolfgang Walz (eds.)
1995 | 316 Pages | ISBN: 0896033112 | PDF | 24 MB
E. Neher and B. Sakman were the first to monitor the opening and closing of single ion channels and membranes by conductance measurements. In 1976, they used firepolished micropipets with a tip diameter of 3-5 pm to record currents from a small patch of the membranbe of sk- etal muscles, thereby decreasing background membrane noise. In order to reduce the dominant source of background noise-the leakage shunt under the pipet rim between m- the muscle membrane had to be treated brane and gla- enzymatically. Despite these early limitations, a new te- nique was born -the patch-clamp technique. The final bre- through came in 1981 when the same authors, in collaboration with 0. P. Hamill, A. Marty, and F. J. Sigworth, developed the gigaohm seal. Not only did this improve the quality of recordings, it was now possible to gently pull the membrane patch with the attached pipet off the cell and study its trapped ion channels in isolation. Another offshoot of the gigaohm seal technique was the whole-cell patch-clamp technique, in which the patch is ruptured without breaking the seal. This technique is really a sophisticated voltage-clamp technique and also allows for the altering of cytoplasmic constituents if the experimenter so wishes. The first part of Patch-Clamp Applications and Protocols presents modern developments associated with the techn- ogy of patch-clamp electrodes, of cell-free ion channel reco- ing, and of the whole-cell patch-clamp technique.

Pain Research: Methods and Protocols By Z. David Luo (auth.), Z. David Luo MD, PhD (eds.)
2004 | 302 Pages | ISBN: 1588291030 | PDF | 4 MB
Despite significant advances in molecular biology techniques and in our understanding of the physiology and the behavioral pharmacology of pain transduction, effective, target-specific therapeutic agents for chronic pain are still lacking. In Pain Research: Methods and Protocols, leading researchers who have first-hand experience describe, in step-by-step detail, diverse and novel techniques for dissecting the molecular mechanisms of pain transduction. These readily reproducible methods employ a variety of multidisciplinary approaches ranging from animal pain models and single neutron selection to in vitro single-cell mRNA amplification. The collection includes not only standard, cutting-edge methods, but also novel techniques only recently applied to pain research. The protocols follow the successful Methods in Molecular Biology™ series format, each one offering step-by-step laboratory instructions, an introduction outlining the principle behind the technique, lists of equipment and reagents, and tips on troubleshooting and avoiding known pitfalls. Versatile and easy-to-use, Pain Research: Methods and Protocols offers today's pain researchers in academic and pharmaceutical laboratories powerful tools to unravel the cellular and molecular complexity of pain transduction and set the stage for the next generation of pain medications.

New-Technology Flowmeters: Volume I
English | 2023 | ISBN: 036765542X | 189 Pages | PDF (True) | 10 MB
New-Technology Flowmeters describes the origin, principle of operation, development, advantages and disadvantages, applications, and frontiers of research for new-technology flowmeters, which include Coriolis, magnetic, ultrasonic, vortex, and thermal. Focusing on the newer, faster growing flowmeter markets, the book places them in the context of more traditional meters such as differential pressure, turbine, and positive displacement. Taking an objective look at the origins of each flowmeter type, the book discusses the early patents, for each type, and which companies deserve credit for initially commercializing each flowmeter type. This book is designed for personnel involved with flowmeters and instrumentation, including product and marketing managers, strategic planners, application engineers, and distributors.
Neuropeptide Protocols By J. Michael Conlon (auth.), G. Brent Irvine, Carvell H. Williams (eds.)
1997 | 386 Pages | ISBN: 0896033996 | PDF | 8 MB
After 20 years of intensive effort, novel neuropeptides continue to be discovered, and the field of neuropeptide research is still expanding. As new analytical techniques become available, their applicability to the study of neu ropeptides brings fresh insights into the properties and functions of these ubiq uitous chemical messengers. Presented in this single volume, Neuropeptide Protocols, are 33 chapters covering these new techniques, together with more established methods. Each contributor is actively engaged in neuropeptide research and so brings to his or her description an awareness of the practical problems inher ent in the method, and provides sound advice on how to overcome them. The format conforms to the style of previous books in the Methods in Molecular Biology series. Each chapter provides an instruction to the technique, and item ized list of equipment and reagents, and a step-by-step set of instructions to enable practitioners to reproduce the method. The Notes section gives insights into pitfalls or critical stages, tips to overcome these obstacles, and sugges tions for extensions or modifications of the basic protocol. Neuropeptide Protocols is intended as a benchtop manual providing the entire gamut of techniques that form the essential tool kit of the practicing neuropeptide researcher. It will be useful for those new to the field, as well as for established workers who wish to try a new technique for the first time.
